Instrumentation and Techniques

The iBiOs provides access to eleven advanced fluorescence microscopy systems. Three confocal laser scanning microscopes are available, out of which one is equipped with a Ti:Sa laser for non-linear optical (NLO) imaging (Zeiss LSM 510 META NLO) and another is equipped with time-correlated single photon counting for fluorescence lifetime imaging (FLIM) and fluorescence correlation techniques for protein interaction analysis by FRET and cross-correlation (Olympus LSM Fluoview). The latter system was recently upgraded with an anisotropy option to quantify protein dimerization and oligomerization by homo-FRET. A turn-key wide-field deconvolution microscopy system is accessible for fast 3D multicolor imaging (DeltaVision Elite). For the application of various SMLM techniques, three total internal reflection fluorescence microscopes (TIRFM) equipped with powerful laser systems for PALM and STORM applications are available with multicolor detection (Olympus TIRF 3-Line, Olympus TIRF 4-Line SMT, Olympus TIRF 4-Line STORM). Another TIRF microscope combines STORM imaging with an atomic force microscope for correlative approaches (Olympus TIRF 1-Line with JPK AFM). By the integration of a powerful NIR laser into a fluorescence wide-field microscope, up-conversion nanoparticle imaging can be performed (UCNP Setup). A dual camera spinning disk system is the method of choice for fast volumetric fluorescence imaging of living cells and tissues under moderate bleaching conditions and resolution (Zeiss Cell Observer SD). The Lattice Light-Sheet Microscope enables high resolution 3D live cell imaging under very low bleaching conditions and its very thin light-sheets can be used for SMLM inside cells, tissues or small organisms.
