Instrumentation and Techniques

The iBiOs provides access to fourteen advanced fluorescence microscopy systems. Five confocal laser scanning microscopes with different features are available. The Leica SP5 is a routine cLSM with live cell periphery and sensitive GaAsP detectors. Two cLSMs are equipped with a femto-second laser sytem for non-linear optical (NLO) imaging and photomanipulation. The Zeiss LSM 510 META NLO is a routine up-right microscope for multi-photon excitation applications, whereas the Olympus LSM FLUOVIEW 3000 NLO is a multi-modal imaging platform with tailored design for upconversion and harmonic nanoparticles. Another cLSM is equipped with time-correlated single photon counting for fluorescence lifetime imaging (FLIM) and fluorescence correlation techniques (Olympus LSM FlUOVIEW 1000). Our Zeiss LSM 880 with Airyscan detector enables 1.7x higher spatial resolution by image scanning microscopy.
With respect to camera-based wide-field imaging, we provide a turn-key deconvolution microscopy system for fast 3D multicolor imaging (DeltaVision Elite). For the application of various SMLM techniques (single molecule tracking, PALM, STORM, PAINT), three total internal reflection fluorescence microscopes (TIRFM) are available (Olympus TIRF 3-Line, Olympus TIRF 4-Line SMT, Olympus TIRF 4-Line STORM). Another TIRF microscope combines STORM imaging with an atomic force microscope for correlative approaches (Olympus TIRF 1-Line with JPK AFM). By the integration of a powerful NIR laser into a fluorescence wide-field microscope, up-conversion nanoparticle imaging can be performed (UCNP Setup). A dual camera spinning disk system is the method of choice for fast volumetric fluorescence imaging of living cells and tissues under moderate bleaching conditions and resolution (Zeiss Cell Observer SD). Our second spinning disk system CQ1 is a fully automated benchtop high-content screening system. The most advanced light microscope at iBiOs is the Lattice Light-Sheet Microscope enabling high resolution 3D live cell imaging under very low bleaching conditions and highest temporal resolution. With its very thin light-sheets, SMLM can be permformed inside cells, tissues or small organisms.
